Germination test is a parameter for evaluation of the physiological quality of seeds, however, for most of the native forest species there is still a great lack of information about the germination test, which makes it difficult to elaborate protocols for evaluation the physiological quality. This study aimed to define the ideal temperature and substrate for germination test of Cassia leptophylla seeds. The experimental design was completely randomized in a 4x4 factorial with four substrates (sand, blotting paper, paper towel and vermiculite) and four temperatures (20, 25, 30 and 20-30 ºC) with four replicates of 50 seeds. The variables analyzed were: normal seedlings, first germination count, germination speed index and mean germination time. The germination test can be performed on sand substrate at 30 ºC and paper towel at the alternating temperature of 20-30 ºC.
